Very little is known about SMRPG2 yet. It was shown in brief chunks on a video at Nintendo's Space World show. Very little was said about it by Miyamato at the press conference. He said only that it will be in Yoshi's Story's 2 1/2 D gameplay and will be geared at younger kids. The screen shots look reminiscent of the rendered cut sceens in the original SMRPG. Whether these are cuts from the new game, or actual gameplay, isn't known. The main thing this video showed was that the game is started and on it's way to a 64DD near you (eventually).
